Plant suspension cell cultures display many features of the innate immune responses observed in planta and have been extensively applied to the study of basal and race-specific defences. However, no single model including photosynthetic cultured cells has been used for the exhaustive characterization of both basal and race-specific defences to date. In this article, we report the activation of basal and race-specific defences in green cultured cells from Arabidopsis thaliana . Inoculation of cultured cells with isogenic virulent or avirulent strains of Pseudomonas syringae pv. tomato DC3000 (Pst) was used to evaluate race-specific defences. The proliferation of avirulent Pst was found to be lower than that of virulent Pst in the inoculated cultures. Extracellular pH changes, sustained oxidative burst (5–13 h post-inoculation), enhancement of salicylic acid, and massive cell death were specifically stimulated by the avirulent bacterium. Neither avirulent nor virulent Pst induced markers of basal resistance, such as callose deposition or early oxidative burst (1–5 h post-inoculation). However, both basal defences were activated when cells were exposed to Pseudomonas syringae pv. phaseolicola or to the Pst mutant defective in the type III secretion system (TTSS), Pst-hrpL. Thus, in these cells, basal defences may be inhibited by Pst in a TTSS-dependent manner. Recapitulation of classical defence features demonstrates the usefulness of this system for the fine characterization of plant innate immune components.  相似文献

SUMMARY Pneumocystis carinii pneumonia (PCP) is usually diagnosed by examination of BAL, a sample often unpleasant to be collected from immunocompromised host affected by acute respiratory disease. We studied by the Internal Transcribed Spacers (ITSs) nested PCR the presence of P. carinii DNA in serum and Peripheral Blood Mononuclear Cells (PBMC) during acute episodes of PCP to test blood as a possible noninvasive diagnostic tool.  相似文献

A purified (Na+ + K+)-ATPase large subunit obtained from microsomes by water-alcohol extraction was incorporated into a bilayer lipid membrane. The protein formed in the membrane conductance channels which were sensitive to ouabain and selective for monovalent cations. ATP activated these channels in the presence of sodium and potassium ions. When sodium ions were eliminated ATP did not change the conductance of the modified membrane whereas p-nitrophenyl phosphate increased it. The (Na+ + K+)-ATPase large subunit incorporated into bilayer lipid membrane possessed an ATPase activity. The presence of a potential on the membrane was a necessary condition for the enzyme incorporated into a bilayer lipid membrane to show high ATPase activity. Increasing the potential above 100 mV resulted in the closing of conductance channels.  相似文献

In the present study, we employed three mitochondrial DNA genetic markers in a phylogenetic analysis aiming at the delineation of the relationships amongst nominal Trachelipus kytherensis populations, as well as between populations of this species and of Trachelipus aegaeus and a new form, occurring syntopically with the latter. Both the phylogenetic analysis and the genetic distances separating populations, show the presence of several distinct and well differentiated clades that undermine the monophyly of T. kytherensis. On the other hand, despite the insular distribution of T. aegaeus populations, their divergence is low and the monophyly of this taxon can be rescued by the inclusion of two more insular populations previously assigned to T. kytherensis. The patterns of genetic divergence among clades are only partially congruent with the geographic distribution of populations. The validity of taxonomic characters used so far in the genus appears to be questionable; therefore, a more comprehensive phylogenetic study at a population level is deemed necessary for understanding the divergence of Trachelipus lineages. © 2008 The Linnean Society of London, Biological Journal of the Linnean Society, 2008, 95 , 361–370.  相似文献

From a sandy beach in southern Chile (Quellón Viejo, Chiloé Island) five species of the nematode family Epsilonematidae are described or redescribed: Bathyepsilonema spongiosum sp.n., Epsilonema dentaturn Lorenzen, 1973, E. serrulatum Lorenzen, 1973, Metepsilonema chilotum sp.n. and M. leptaleum Lorenzen, 1973. In the studied beach Epsilonematidae represent up to 33% of the interstitial nematodes. Epsilonema dentaturn is the most common species; in most of the samples, it represents more than 50% of all Epsilonematidae found. The epsilonematids occur mainly in a narrow zone at the midtidal level.  相似文献

We established a cytochrome b (cyt b ) phylogeny for six species of social voles. A Bayesian approach to phylogenetic reconstruction (BI) and a maximum likelihood (ML) tree revealed a dichotomy into two major clusters, namely a Microtus guentheri cluster and a M. socialis cluster. The three main lineages that emerged within each of these two clusters were separated by the K2P divergences which are above the intraspecific variation in Microtus . All six species were also retrieved in the minimum spanning network. Within its present taxonomic scope, M. guentheri is paraphyletic and consists of two allopatric sibling species: M. guentheri (Syria, Israel) and M. hartingi (Anatolia and the Balkans). The closest relative to these two species is M. dogramacii , which is possibly a sister species to M. hartingi . The two geographic samples were identified as M. irani , one from Shiraz (Iran) and the other from Balkusan (Turkey). The cyt b sequence confirmed the specific status of M. anatolicus within the M. socialis cluster. Although five species of social voles occur within a radius of < 500 km in the north-eastern corner of the Mediterranean, small-scale sympatry is exceptional. Abstract. Scanning electron microscopy (SEM) was used to depict and describe the eggs of five species of phlebotomine sandfly from the Andean region of Peru: Lutzomyia caballeroi, Lu.noguchii, Lu.peruensis, Lu.tejadai and Lu.verrucarum. Two new types of chorionic sculpturing of sandfly eggs are reported: these were named disperse (Lu.tejadai) and verrucose (Lu.noguchii). The aeropylar area of the eggs is also described for the first time for neotropical sandflies. These character states appear to vary for ecological rather than phylogenetic reasons and could be used for species identification.  相似文献

A novel, unbiased approach to plant viral disease diagnosis has been developed which requires no a priori knowledge of the host or pathogen. Next-generation sequencing coupled with metagenomic analysis was used to produce large quantities of cDNA sequence in a model system of tomato infected with Pepino mosaic virus . The method was then applied to a sample of Gomphrena globosa infected with an unknown pathogen originally isolated from the flowering plant Liatris spicata . This plant was found to contain a new cucumovirus, for which we suggest the name 'Gayfeather mild mottle virus'. In both cases, the full viral genome was sequenced. This method expedites the entire process of novel virus discovery, identification, viral genome sequencing and, subsequently, the development of more routine assays for new viral pathogens.  相似文献

The anatomy of the Hydrocharitaceae is described in relation to the modes of life of the family. Leaf morphology depends on the ecology of the species. The xylem is simple with relatively primitive characters, but the phloem shows a contradictory evolutionary trend, possessing the highly evolved characters of large, wide sieve-tube elements with more or less horizontal, simple sieve-plates and a single companion cell to each element. Characters of taxonomic value within the family exist but are lewer and less satisfactory than those in families such as the Cyperaceae; such characters are the shape of the axillary scales, the pattern of occurrence of tannin secretory cells and the diaphragm cells of roots.
